OGR1 inhibited MCF7 breast cancer cell migration through a Gα 12/13 -Rho-Rac1 pathway. Cells were pretreated with the solvent or PTX (1 μM) for 16 h, or transfected with the RGS plasmid for 48 h. (A) Cell migration was analyzed by transwell migration assays after treatment or using transient transfected cells 72 h post-transfection. ***P<0.001. (B) and (C) The protein activation levels were examined by pull-down and Western blot analyses as described in Methods. Rho, Rac1, Cdc42 and p115 RGS,as well as α-tubulin were analyzed in whole cell lysates. Representative results are from three independent experiments.
